New procedure variant tomorrow such that I desperately need to sit down and figure it out and make the grepping buffers so of course the boss hands me a long list of other things needing doing. Including asking me to finish up his yeast transformation on the way out the door to go play squash or something. Okay, I was ridiculously late this morning, but still.
Here's what I absolutely need to get done:
feed the transfects (fast)
finish the yeast transformation (maybe 30 min?)
reread the methods paper (let's say 20 min, very optimistically)
make buffers (30 min? an hour? will only know once I've reread the paper)
pick up the rest of my developed films and develop the long exposure
Here's what else I should get done that the boss considers a priority:
picking colonies (about 20 min except I need to make LB first)
Also in theory I was supposed to gel-purify the fragments for subcloning and set up ligations. Hah.
But, basically, will be here the rest of the night.
This is your cue to send me fun/entertaining/supportive email so at least I have something to look forward to. Or somebody write me 200 words of Lance love or something.
